SH3BP2 (3BP2) is involevd in NK-activation. 3BP2 is coupled to activating receptors on NK cells.It is phosphorylated after NK stimulation.
CD244 ligation induces 3BP2 phosphorylation and Vav recruitment.
The adaptor protein 3BP2 binds human 2B4 (CD244) and links this receptor to Vav signaling, PLCG, ERK activation, and NK cell killing.
SH2 domain of 3BP2 is required for optimal tyrosine phosphorylation of 3BP2 following FcR cross-linking and for its ability to associate with the transmembrane adaptor protein LAT.
Phosphorylated tyrosine-183 of 3BP2 binds PLCG1 and PLCG2 and Vav1 during activation of NK cells through natural cytotoxicity receptors.
