KIRs constitute a family of slightly polymorphic receptors with distinct MHC-I-binding profiles for classical HLA-A, HLA-B, and HLA-C molecules. Most KIRs with two Ig domains (KIR2DL) recognize subsets of the HLA-C allotypes.
KIR2DL3 inhibits NK cells downstream of HLA-C via SHP-1 binding and activation.

KIR2DL3, KIR2DL1, KIR2DS4, KIR3DL1, KIR3DL2, KIR2DL4 interact with SHP-2 in NK cells
and probably are phosphorylated by LCK 5directly demonstrated for KIR2DL3 only.
