The ITAM can be part of the same polypeptidic chain that engages the ligand, as in the case of Fc??RIIA and Fc??RIIC, or of a separate ?? subunit that associates noncovalently with the ligand-binding subunit of the receptor, as in the case of Fc??RI and Fc??RIIIA. Upon receptor clustering, the ITAM associates with and becomes phosphorylated by Hck, Lyn (33, 34), and/or Fgr (35).

In neutrophiles Fgr is physically and functionally associated with Fc gamma RII and involved in Fc gamma RII-mediated signal transduction pathways.
