Through paracrine signaling molecules, TGF-beta and IL-1beta, cancer cells activate stromal fibroblasts and induce the expression of fibroblast activation protein (FAP). FAP, in turn, affects the proliferation, invasion and migration of the cancer cells. We report that TGF-beta and IL-1beta are important factors in inducing differentiation of myofibroblasts and expression of functional markers, notably alpha-SMA.

IL1 signaling has both antitumor effect (low dose of IL B and menbran associated IL1A) and protumor effect (high dose of IL1) in tumor microevironment,

Microenvironmental IL-1 beta and, to a lesser extent, IL-1 alpha are required for in vivo angiogenesis and invasiveness of different tumor cells.

Tumor-derived IL-1?? secreted into the tumor microenvironment has been shown to induce the accumulation of MDSC that promotes tumor growth.

IL-1?? activates MDSC in vitro and in vivo through an IL-1RI/NF-??B pathway.
mRNA expression of several NF-??B target genes such is IL6, IL1B,TNF are IL1B/NFkB dependent in MSDC

IL-1R???deficient mice have a delayed accumulation of MDSC and reduced primary and metastatic tumor progression. Accumulation of MDSC and tumor progression are partially restored by IL-6, indicating that IL-6 is a downstream mediator of the IL-1?????induced expansion of MDSC. IL-1Ra (IL-1R antagonis) ???/??? MDSC are more suppressive than BALB/c MDSC.
