Cytosolic DNA induces type I IFNs through the endoplasmic reticulum membrane protein STING, which subsequently activates the transcription factors NF-??B and IRF3 through the kinases IKK and TBK1, respectively.
As a high-affinity ligand for STING, 2???3???-cGAMP binds and activates STING through a series of structural changes. Activated STING then recruits TBK1 to phosphorylate IRF3 and activate IKK to phosphorylate I??B??, leading to I??B?? degradation.
