In immature DCs, internalization of MHC class II molecules from the plasma membrane may require the ubiquitin ligase MARCH1, which is controlled by interleukin-10 (Measurement of??March1??mRNA by quantitative PCR in these DC cultures demonstrated that IL-10 increased??March1??mRNA by approximately sixfold).
CD83 on mature DCs prevents this ubiquitylation of MHC class II molecules and thus stabilizes MHC class II molecules on the cell surface. Addidionally it prevents CD86 ubiquitinqtion by MARCH1.
